Lab 2a
Purpose: What are positive indicator tests for proteins, carbohydrates, and fats? Do parts of an egg test positive, for protein, carbohydrate, and/or fat?
Materials: Eggs Tubes, glass, 13x100 mm Sodium hydroxide
Beaker, 250 mL Peg racks Cupric sulfate 5-hydrate
White Vinegar Glucose Oil
Plastic Wrap Benedict's solution Sudan IV Solution
Slotted Spoon Hot plate stirrer Scalpel Handles #4
Distilled water Soluble starch Scalpel Blades #22
Beaker, 100 mL Lugol's Iodine Solution Plastic trays
Pipets, 5 mL Vortex mixer
Pipet Pump Gelatin
Procedure: 1. Place an uncooked egg into a beaker of white vinegar. Label and cover it with plastic wrap. Leave it for 24 to 48 hours.
2. After 24 hours gently pick up the egg with a slotted spoon and rinse it with water to remove the vinegar.
3. Feel the outer membrane of the cell. It is thin and flexible. Place the egg in a beaker of 5% NaCl solution for 24 hours. Describe the appearance of the egg after 24 hours.
4. Gently rinse the egg with water. Place the egg in a beaker of distilled water for 24 hours.
5. Gently slice oped the egg membranes and let the egg white dripthrugh the slot into the 100 mL beaker. (do not peirce yolk)
6. Place yolk into another beaker
PART 2
MONOSACCHARIDE INDICATOR STANDARD TEST
1. Test for glucose: in a test tube, mix 2 ml of a 2% glucose solution with 2mL of Benedict's solution. Heat for 2 min in boiling hot water bath. Record all color changesand lenght of time for color to appear.
2. Test for water: In a test tube, mix 2 mL of water with 2 mL of Benedicts solution. Heat for 2 min in boiling hot water bath. Record all color changesand lenght of time for color to appear.
STARCH INDICATOR STANDARD TEST
1. Test for starch: In a test tube, mix 2mL of well-mixed starch suspension with 0.25ml of Lugols iodine.Genly swirl to mix. Record color change.
2. Test for water: In a test tube, mix 2 mL of water with 0.25ml of Lugols iodine. Genly swirl to mix. Record color change.
PROTEIN INDICATOR TEST
1. Test for protein. Place 2mL of gelatin solution in a test tube. add 0.5 mL of 10% NaOH and CuSO4 and gently vortex to mix well and record color change.
2. Test for water. place 2 mL of deionized water in a test tube. add 0.5 mL of 10% NaOH and CuSO4 and gently vortex to mix well and record color change.
PART 3
Test each of the egg components for the prsence of moma saccarides starch protein and lipids
1. Conduct each indicator test as described in part 2, but substitute each egg component to be tested for sugar starch protein or fat in the test. All volumess and test conditions are the same as in part 2: only the item tested is changed.

CONCLUSION
During this lab we tested parts of the egg for several types of molecules. Positive indicator tests for protein are when the substance turns purple. Positive indicator tests for carbohydrates proved positive with quick blunt color change. Positive indicator fats result in a decent change in color.(for data see tables) One down fall of this experiment was that we may have accidentally mixed pipettes. This could have resulted in different results because some yolk could have gotten in the test tube when we only wanted the white. However, our results did match most of our fellow scientists and it matched what we had hypothesized. One different experiment of extension of this experiment could be doing it again with different eggs and comparing the results with the chicken egg. We could also use indicator test to test for different things in your systems like drugs or diseases.
REFLECTION
One thing that I liked about this experiment was that it was very interesting and new, something I had never done or seen before. Also I liked that we did many different experiments in this one lab and we weren't done in just a few minutes. The fact that we had to wait overnight made me that much more curious. Me and my partner worked very well together. We shared the work load and got along great. I thin that the lab was very well designed and we executed it well. I would honestly change nothing about this lab.
